This is a working overview of Melanotan II, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-01-17 and is reviewed periodically as new material appears.
Regulatory treatment varies by country. In the United States, melanotan-2 is not approved for any indication, and products marketed for human use fall outside the approved drug framework. Some other jurisdictions have placed it under prescription controls or listed it as a prohibited or restricted substance. Online listings frequently describe the material as a research chemical, a category that does not carry the same manufacturing and labelling requirements as approved medicines.
Solid peptide material is generally stable when kept cold and dry. Common practice is storage at -20 degrees Celsius or lower, with desiccant and protection from light. Repeated freeze-thaw cycles and exposure to moisture are associated with degradation, aggregation, or loss of material. Once dissolved, stability depends on solvent, concentration, and temperature, and solutions are usually treated as short-lived unless stability data support longer periods. Handling notes typically emphasise minimising time at ambient temperature.
Identity and purity are assessed with chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the target peptide from related impurities and degradation products, and the resulting retention time is compared against a reference standard. Mass spectrometry, often coupled to liquid chromatography, confirms molecular mass. Amino acid analysis or peptide mapping can provide additional sequence-level confirmation when required. Results are only as reliable as the reference materials used alongside them.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.

Regulatory status differs by country, and in many places supplying the compound for human consumption is unlawful. Vendors frequently label material as intended for research use only, a designation that shifts stated purpose but does not create a legal pathway for personal use. Certificates of analysis accompanying such products vary widely in detail and provenance. Third-party testing exists but is voluntary, and results are rarely linked to a specific lot in a publicly verifiable way.
Identity testing for a synthetic peptide relies on several complementary methods. Reversed-phase high-performance liquid chromatography separates the target from related impurities and reports purity as a percentage of total peak area. Mass spectrometry confirms molecular mass and can reveal deletions or truncations. Amino acid analysis and peptide mapping provide sequence-level confirmation, while counter-ion content and residual solvents are measured separately. A purity figure alone does not establish identity, so a complete dataset combines chromatographic and spectrometric evidence.
Lyophilized peptide powder is generally stored frozen, protected from light and moisture. Tryptophan residues are susceptible to oxidation, and the lactam bridge can hydrolyze under strongly acidic or basic conditions. Solutions prepared for laboratory work degrade faster than dry powder, and repeated freeze-thaw cycles accelerate loss. Common practice is to aliquot solutions before freezing and to avoid alkaline buffers. Reported stability windows vary with concentration, buffer, and temperature, so exact shelf lives are method-specific rather than universal.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
=== UK 1947–1991 === Shortly after being named in testimony before the House Un-American Activities Committee, the Eltentons returned to England. He first took a senior position at the Shell physics research laboratory, but after investigation by MI5 was moved to an area solely concerned with refinery operations.
== Published works == Rowland has published over 270 research articles in international peer-reviewed journals, and over 60 book chapters and conference reports. He co-authored with Thomas Tozer two standard textbooks: Clinical Pharmacokinetics and Pharmacodynamics: Concepts and Applications, now in its 5th edition, and Essentials of Pharmacokinetics and Pharmacodynamics.
The delayed time to peak levels (2–3 hours) and long elimination half-life (12 hours) of suvorexant are less than ideal for an insomnia medication as they result in a delayed onset of effect and significant next-day side effects such as daytime sedation. Orexin receptor antagonists with shorter half-lives and faster onsets of action are theoretically more optimal for therapeutic use as sleep aids. The ideal insomnia medication would not have a duration of action extending beyond about 8 hours. Relative to suvorexant, daridorexant has a shorter half-life (8 hours) while lemborexant has a longer half-life (17–55 hours). However, although lemborexant has a longer terminal elimination half-life than suvorexant, it appears to be more rapidly cleared in the earlier phases of elimination. The investigational agents seltorexant and vornorexant, which are still in clinical trials, have comparatively very short half-lives in the range of 1.5 to 3 hours.
== Revisionism == The role of the United States in the Vietnam War disillusioned New Left historians and created many historians with sympathy towards the Viet Cong communist position and antipathy towards American policies. Much more important were the revisionists who argued that both United States and the Soviet Union were responsible for blundering into the war and rejected the premises of "containment". They battled the "orthodox" historians. "Revisionist" accounts emerged in the wake of the Vietnam War in the context of a larger rethinking of the United States role in international affairs, which was seen more in terms of American empire or hegemony. In the specific context of the Vietnam War, this "revisionist" narrative became the historiographical orthodoxy, whereas revisionism in Vietnam War historiography came to defend the US policy of containment. While the new school of thought spanned many differences among individual scholars, the works comprising it were generally responses in one way or another to William Appleman Williams 1959 volume, The Tragedy of American Diplomacy. Williams challenged the long-held assumptions of "orthodox" accounts, arguing that Americans had always been an empire-building people even while American leaders denied it. The influence of Williams, who taught at the University of Wisconsin–Madison, and several of his students who subsequently published works on these themes, was enough to create what became known as the Wisconsin School of American diplomatic history.
=== Alternative medicine === An experimental treatment, enzyme potentiated desensitization (EPD), has been tried for decades but is not generally accepted as effective. EPD uses dilutions of allergen and an enzyme, beta-glucuronidase, to which T-regulatory lymphocytes are supposed to respond by favoring desensitization, or down-regulation, rather than sensitization. EPD has also been tried for the treatment of autoimmune diseases, but evidence does not show effectiveness. A review found no effectiveness of homeopathic treatments and no difference compared with placebo. The authors concluded that, based on rigorous clinical trials of all types of homeopathy for childhood and adolescent ailments, there is no convincing evidence that supports the use of homeopathic treatments. According to the National Center for Complementary and Integrative Health, US, the evidence is relatively strong that saline nasal irrigation and butterbur are effective, when compared to other alternative medicine treatments, for which the scientific evidence is weak, negative, or nonexistent, such as honey, acupuncture, omega 3's, probiotics, astragalus, capsaicin, grape seed extract, Pycnogenol, quercetin, spirulina, stinging nettle, tinospora, or guduchi.

Arrestins block GPCR coupling to G proteins in two ways. First, arrestin binding to the cytoplasmic face of the receptor occludes the binding site for heterotrimeric G-protein, preventing its activation (desensitization). Second, arrestin links the receptor to elements of the internalization machinery, clathrin and clathrin adaptor AP2, which promotes receptor internalization via coated pits and subsequent transport to internal compartments, called endosomes. Subsequently, the receptor could be either directed to degradation compartments (lysosomes) or recycled back to the plasma membrane where it can again signal. The strength of arrestin-receptor interaction plays a role in this choice: tighter complexes tend to increase the probability of receptor degradation (Class B), whereas more transient complexes favor recycling (Class A), although this rule is far from absolute. More recently direct interactions between Gi/o family G proteins and Arrestin were discovered downstream of multiple receptors, regardless of canonical G protein coupling. These recent findings introduce a GPCR signaling mechanism distinct from canonical G protein activation and β-arrestin desensitization in which GPCRs cause the formation of Gαi:β-arrestin signaling complexes.
Absorption is the journey of a drug travelling from the site of administration to the site of action. The drug travels by some route of administration (oral, topical-dermal, etc.) in a chosen dosage form (e.g., tablets, capsules, or in solution). Absorption by some other routes, such as intravenous therapy, intramuscular injection, enteral nutrition, is even more straightforward and there is less variability in absorption and bioavailability is often near 100%. Intravascular administration does not involve absorption, and there is no loss of drug. The fastest route of absorption is inhalation. Absorption is a primary focus in drug development and medicinal chemistry, since a drug must be absorbed before any medicinal effects can occur. Moreover, the drug's pharmacokinetic profile can be easily and significantly changed by adjusting factors that affect absorption.
Bruce Glick grew up in Pittsburgh, Pennsylvania and was interested in birds as a child. His father, Peter Glick, was the Secretary of Labor for Pennsylvania. Glick served in World War II. He went to Rutgers University and studied birds majoring in poultry science, graduating in 1951. In 1950 he married Kay McCall. He received an M.S. degree from the University of Massachusetts in genetics in 1952 and attended Ohio State University as a Ph.D. student, graduating with a PhD in physiology in 1955. While there, he worked on determining the purpose of the Bursa of Fabricius, a gland that he was able to remove from a goose without any apparent effect. A fellow graduate student, Timothy Chang, worked with Glick's geese in a different study, and noticed that the birds without the Bursa of Fabricius did not produce expected antibodies. Glick and Chang wrote up the results of this study and were unable to get it published in Science, so it was published in Poultry Science in 1956. Their publication, considered a landmark paper, is one of the most cited works from Poultry Science.
== Boron-8 == Boron-8 is an isotope of boron that undergoes β+ decay to beryllium-8 with a half-life of 771.9(9) ms. It is the strongest candidate for a halo nucleus with a loosely-bound proton, in contrast to neutron halo nuclei such as lithium-11. Although boron-8 beta decay neutrinos from the Sun make up only about 80 ppm of the total solar neutrino flux, they have a higher energy centered around 10 MeV, and are an important background to dark matter direct detection experiments. They are the first component of the neutrino floor that dark matter direct detection experiments are expected to eventually encounter.
The alkaloid product of the reaction, neopinone, can spontaneously isomerise to the next intermediate, codeinone, in the pathway to morphine via codeine but this step is catalysed in opium poppy by the enzyme neopinone isomerase (NISO).

Peña and Javier Milei met on 9 December 2023 and agreed to "deepen" Mercosur. Peña stated that he had placed himself "at Milei's disposal" and attended his presidential inauguration in December 2023. Paraguay's vice minister of commerce, Óscar Stark, stated on 13 December 2023 that the economic policy of the new Argentine government, which aspires to eliminate multiple exchange rates and move toward exchange-rate unification in the short term, works against the smuggler's business and is therefore favorable for Paraguay, and that "it is going to be much easier to talk than with the previous government".
== Role in winemaking == The primary role of malolactic fermentation is to deacidify wine. It can also affect the sensory aspects of a wine, making the mouthfeel seem smoother and adding potential complexity in the flavor and aroma of the wine. For these other reasons, most red wines throughout the world (as well as many sparkling wines and nearly 20% of the world's white wines) today go through malolactic fermentation. Malolactic fermentation deacidifies the wine by converting the "harsher" diprotic malic acid to the softer monoprotic lactic acid. The different structures of malic and lactic acids leads to a reduction of titratable acidity (TA) in the wine by 1 to 3 g/L and an increase in pH by 0.3 units. Malic acid is present in the grape throughout the growing season, reaching its peak at veraison and gradually decreasing throughout the ripening process. Grapes harvested from cooler climates usually have the highest malic content and have the most dramatic changes in TA and pH levels after malolactic fermentation.
As of 2013, 19,400 people inhabited the Kuril Islands, of which 16,700 lived on the four disputed southern islands and 2,600 lived on Paramushir, the northernmost large island; the islands in between are uninhabited. These include ethnic Russians, Ukrainians, Belarusians, Tatars, Nivkhs, Oroch, Japanese and Ainus. Russian Orthodox Christianity is the main religion. Some of the villages are permanently occupied by Russian soldiers. Others are inhabited by civilians, who are mostly fishermen, workers in fish factories, dockers, and social sphere workers (police, medics, teachers, etc.). Construction works on the islands have attracted migrant workers from the rest of Russia and other post-Soviet states. As of 2014, there were only 8 inhabited islands out of a total of 56.
Dyneins, one of the three major classes of motor protein, are AAA proteins which couple their ATPase activity to molecular motion along microtubules. The AAA-type ATPase Cdc48p/p97 is perhaps the best-studied AAA protein. Misfolded secretory proteins are exported from the endoplasmic reticulum (ER) and degraded by the ER-associated degradation pathway (ERAD). Nonfunctional membrane and luminal proteins are extracted from the ER and degraded in the cytosol by proteasomes. Substrate retrotranslocation and extraction is assisted by the Cdc48p(Ufd1p/Npl4p) complex on the cytosolic side of the membrane. On the cytosolic side, the substrate is ubiquitinated by ER-based E2 and E3 enzymes before degradation by the 26S proteasome.

Laboratory confirmation typically combines retention time matching on a chromatographic system with mass measurement. A reference standard of known identity is needed for a meaningful comparison. Sequence-level techniques can add further confirmation.
There is no single harmonised standard governing these products, and manufacturing conditions differ. Impurities may include truncated sequences, oxidation products, residual solvents, and counter-ions. Independent testing is often the only way to characterise a given lot.
Reported studies have looked at pigmentation, photoprotection, and metabolic or appetite-related endpoints in small experimental settings. Much of the evidence base is early-stage and limited in size. Questions about long-term effects and clinical relevance remain open.
It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
